Does 5-HTP Help With Sleep?

The clearest modern human signal comes from a 2024 randomized controlled trial in 30 older adults in Singapore. Participants were assigned to 100 mg of 5-HTP daily or a no-supplement control for 12 weeks. Sleep was tracked using the Pittsburgh Sleep Quality Index and actigraphy, along with serotonin, melatonin and microbiome measures.

The overall result was promising but not universal. The researchers reported favorable changes in certain sleep-quality components, with the clearest subjective improvement among participants who were poor sleepers at baseline. Good sleepers did not show the same pattern. The trial was small, limited to older adults, single-blinded and did not use a placebo control, so it should not be treated as proof that 100 mg of 5-HTP will solve insomnia in the general population.

What About Mood, Anxiety or Depression?

This is where marketing often outruns product-specific evidence. Health Canada’s current 2024 ingredient monograph recognizes “healthy mood balance” at 150–300 mg of 5-HTP per day. Biogénique’s verified physical label, however, directs adults to take 100 mg once daily and licenses the product as a sleep aid and to help relieve nervousness. We therefore do not present “healthy mood balance” as the licensed claim of this specific 100 mg product based only on newer monograph language or website copy.

More importantly, 5-HTP should not be used as a do-it-yourself treatment for depression or anxiety. The current ingredient monograph says not to use 5-HTP with antidepressants and advises health-care consultation for psychological disorders such as anxiety or depression. Health Canada updated its 5-HTP ingredient monograph in August 2024. Ingredient monographs and individual product licences are related but not identical. The monograph now lists healthy mood balance, sleep disturbances associated with mood imbalance and several other ingredient-level uses at defined dose ranges. It also contains stricter safety language, including “do not use if you are taking antidepressants.”
